Abstract: Aceclofenac is a novel NSAID used
to relieve the pain and inflammation associated with
Rheumatoid arthritis, Osteoarthritis and
Ankylosingspondylitis. Fast dissolving tablets
disintegrate or dissolve in saliva or swallowed without
water. As the tablet disintegrates in mouth, this
enhances the clinical effects of drug through absorption
from mouth, pharynx and esophagus leading to an increase
in bioavailability by avoiding first pass liver
metabolism. In the present study solubility was enhanced
by using beta cyclodextrins, sweeteners and flavors were
used to enhance the organoleptic properties of tablets.
Fast dissolving tablets of Aceclofenac were prepared by
direct compression by super disintegrants such as L-HPC,
Crospovidone and S.S.G in different concentrations.All
the formulations were evaluated for the influence of
disintegrates and their concentrations on the
characteristics of Fast dissolving tablets mainly in
terms of disintegration time and dissolution studies.
Tablets containing Crospovidone at 10% concentration
showed 26 sec disintegration time and 93% of drug is
release in 4 min. Tablets containing L-H.P.C AT 10%
showed 35 sec.disintegration time and 98% of drug
release in 2 min. The tablets were also evaluated for
its hardness, friability and other parameters. All
parameters complied with I.P. limits.
Introduction
Fast dissolving tablets are not only indicated for
people who have swallowing difficulties, but also are
ideal for active people. Fast dissolving tablets are
also called as mouth-dissolving tablets, melt-in mouth
tablets, Orodispersible tablets, rapimelts, porous
tablets, quick dissolving etc. Fast dissolving tablets
are those when put on tongue disintegrate
instantaneously releasing the drug which dissolve or
disperses in the saliva. The faster the drug into
solution, quicker the absorption and onset of clinical
effect. Aceclofenac is a novel NSAID used to relieve the
pain and inflammatioassociated with Rheumatoid
arthritis, Osteoarthritis and Ankylosingspondylitis.
The aim & objective of present study is Aceclofenac was
chosen as a drug candidate, since it is widely
prescribed drug especially in elderly patients to
relieve pain and inflammation associated with rheumatoid
arthritis. This fast dissolving dosage forms will be
better accepted by the patients(geriatric).Hence it was
felt that prudent to formulate the drug into fast
dissolving tablet to improve market segment with novel
drug delivery system, the fast dissolving technology was
selected. To objective of current study is To enhance
the drug absorption, To mask the bitter taste of the
drug.
