Licofelone - A Dual COX and 5-LOX Inhibitor

Abstract: Licofelone is the first member of new class of analgesic and anti-inflammatory drugs acting by dual mechanism inhibiting both Cyclooxygenase (COX) & 5-Lipoxygenase (5-LOX). It has been evaluated for the treatment of osteoarthritis (OA), most common form of arthritis. It has been found to be significantly effective in phase III clinical trials completed on patients of osteoarthritis. Regulatory permission for its clinical use is now awaited.
Osteoarthritis, a crippling disorder is becoming an increasingly significant medical problem with the increase of lifespan of man. Presently about 10 % of world's population is suffering from OA. It is a degenerative disease of the joints leading to breakdown of cartilage and proliferation of new bone and connective tissue. So far treatment of OA has been mainly based on symptomatic relief from pain and inflammation by the use of non-steroidal anti-inflammatory drugs (NSAIDS), mainly Ibuprofen & Naproxen, which act by inhibiting COX-2, so preventing the formation of inflammatory prostaglandins but at the same time cause inhibition of COX-I leading to GIT side effects and inhibition of platelet function. This led to development of selective COX-2 inhibitors, celecoxib & others, which spare COX-1 and cause less side effects.
