Design and Evaluation of Pulsatile Drug Delivery System of Metoprolol Tartarate for Effective Treatment of Hypertension.

Introduction: The aim of the present work is to prepare pulsatile release formulation of metoprolol tartarate to increase the therapeutic efficacy of the drug. A core in cup type of tablets are prepared wherein a core tablet an impermeable layer at the bottom and sides of the tablet and soluble hydrophilic polymer at the top are designed. The core tablet contained metoprolol tartarate cellulose acetate propionate is used as impermeable membrane and polyethylene oxide WSR N 80 and polyethylene oxide WSR Coagulant are used as soluble hydrophilic polymers. The top cover layer of soluble hydrophilic polymer is prepared using 32 factorial design. FTIR, DSC studies, quantification of water uptake, top layer expansion, In-vitro dissolution, radial and axial expansion, stereomicroscopic studies short term stability studies are performed. The concentration of the polymer on the top layer influenced the lag time and release pattern of the formulations and increase in the concentration of the polymer increased the lag time and delayed the release where as decrease in the concentration of the polymer decreased the lag time and increased the release rate. The T 70% and T 90% values are also influenced by the concentration of the polymer and its properties. A gel layer at the top of the tablet is observed when it is exposed to dissolution medium because of swelling and the thickness of the gel layer is also influenced by the polymer concentration. The radial and axial expansion studies show a preferential axial expansion of polyethylene oxide. Thus It can be concluded from the study that pulsatile drug delivery system can be formulated which could be useful in chronotherapy of hypertension.
