Studies on Influence of Diluents and Disintegrants on Dissolution of Atorvastatin Calcium Tablets

Introduction: In the present work, an attempt was made to prepare atorvastatin calcium (BCS Class drug) tablets by direct compression with different diluents (Microlac, Avicel, Lactose, Tabletose (100), Dicalcium phosphate and Flowlac) and disintegrating agents (Sodium starch glycolate,Cross povidone,Cross caramelose sodium).

 
The main objective was to study the influence of diluents and disintegrating agents on the dissolution rate of the drug, i.e. Atorvastatin calcium. The physical parameters like weight variation, friability, hardness, content of active ingredient and disintegration time (DT) were also studied. Invitro dissolution studies were carried out in 900ml of 6.8 phosphate buffer as dissolution medium. Most of the tablets complied with the pharmacopoeial standards. Among all the formulations, the tablets containing the diluent Tabletose 100 and among all disintegrating agent cross povidone showed greater dissolution rate in 6.8 phosphate buffer. The dissolution rate of Atorvastatin calcium followed first order kinetics.
