Validated High Performance Liquid Chromatographic Estimation of Voriconazole in Tablet Dosage Form

Abstract
A simple, accurate and reproducible High Performance Liquid Chromatographic method has been developed for the estimation of voriconazole in both pure and pharmaceutical tablet dosage forms. Liquid chromatographic method for the estimation of voriconazole was developed using reversed phase HPLC having ultraviolet detector and separation was carried out on column C18 (250 X 4.6 mm, 5 Micron), using water 486, mode by direct injection. The retention time for standard and test was found to be 7.3 minutes. The method was validated for precision and accuracy. Statistical analysis proved that the method is reproducible and selective for the quantitative estimation of active ingredient in tablet dosage forms and their levels are found to be within the ICH limits.
Introduction
Voriconazole is a broad spectrum antifungal agent from the class of triazoles available for oral use. It is a synthetic fluconazole with little change in the structure where a triazole moiety has been substituted with a flouropyrimidine grouping. It is a fungistatic and is effective against all candida species, including fluconazole-resistant strains, aspergillus species, scedosporium species and fusarium species. Its empirical formula is C16H14F3N5O and molecular weight is 349.3. The drug is not mentioned in any pharmacopoeias, i.e.I.P./B.P./U.S.P. It is officially listed in Martindale. Extensive literature survey revealed that the determination of the drug in pure and dosage forms is not official in any pharmacopoeia and therefore, requires much more investigation. The proposed HPLC method is based on the reversed phase HPLC having UV detector by direct injection based on retention time observation of the drug. Waters HPLC (Model 486) was employed for all the chromatographic measurements. All the chemicals used in the investigation were of analytical grade.
