Taste Masking By Effervescent Method Using Factorial Design Optimization Technique

Introduction: Orodispersible tablets of Promethazine hydrochloride were prepared by effervescent method to mask bitter taste and enhance patient compliance. A 22 factorial design was used to determine the effect of independent variables; croscarmellose sodium and neusilin on the dependant variable in vitro disintegration time. Croscarmellose sodium was used as superdisintegrant and mixture of sodium bicarbonate and citric acid served as effervescent medium along with other standard tablet excipients to improve palatability and mouth feel. The orodispersible tablets were evaluated for hardness, thickness, friability, drug content, wetting time, water absorption ratio, in vitro disintegration time and in vitro drug release. Taste masked formulation with better mouthfeel, improved palatability and faster in vitro disintegration of 2.8 s was selected and subjected for stability studies. To check the validity of the polynomial equation obtained by using optimization technique, a comparison was made between predicted values and experimental values for in vitro disintegration time and correlation plot was obtained. Both the independent variables; croscarmellose sodium and neusilin had significant influence on the dependant variable, in vitro disintegration time. Optimization technique helped in formulating taste masked orodispersible tablets of Promethazine hydrochloride in less time and minimum experimental runs.
