New Analytical Methods for Quantitative Estimation of Eletriptan in Bulk and Pharmaceutical Formulation (M1–M5)

Introduction: Eletriptan is a second-generation triptan drug intended for treatment of headache migraine. Five novel, safe, sensitive, accurate and economical UV Spectrophotometric methods [M1—M5] has been developed and subsequently validated for the determination of Eletriptan in bulk and pharmaceutical formulations. The methods were based on the formulation of colored complex of Eletriptan with different reagents. The absorbance of the formed color complex is measured at the wavelength of maximum absorbance of the complex against the reagent blank. Method M1the acidic dye Tropaeolinooo (TPooo) to form colored species and the absorbance was measured at λmax = 550 nm.Method M2 is based on the reaction of Eletriptan with Folin reagent (FCF) yellowish brown colored species having absorption maxima at λmax =420nm. Method M3 is diazotization of Para nitro Aniline (PNA) with sodium nitrate followed by coupling with drug in alkaline medium (PNA Method) exhibiting absorption maximum at λmax = 490nm. Method M4 is based on the oxidation followed by complexion of drug with 2,2’- Bipyridine (2,2’BPD) in presence of Ferric chloride exhibiting absorption maximum at λmax = 420nm. Method M5 is based on the formation of chloroform extractable ion-association complexes of Eletriptan with Alizarin red S exhibiting absorption maximum at λmax = 600nm. All these methods have different linearity ranges. The developed method was validated according to ICH guidelines and found to be in good accordance with the prescribed values.
