Formulation and Optimization of Nicorandil Floating Pulsatile Pellets By Fluidizied Bed Coating Technique

Introduction: The aim of the work was to formulate Nicorandil floating pulsatile pellets by fluidized bed coating technique where drug loaded pellets were coated with pH dependent polymer eudragit S100 for chronotherapy of angina pectoris which is an underlying cause of heart attack. Nicorandil is categorized as potassium channel opener, water soluble drug and was selected as model drug for treatment of angina pectoris which rely on circadian rhythm. Moreover it has short half-life (1 hour), All these factors contribute to formulate Nicorandil as floating pulsatile pellets These pellets were subjected to evaluation tests like % yield, micromeritic properties, particle size, % entrapment efficiency, % friability, in- vitro floating ability, drug release and kinetics of pellets. The optimized batch exhibited good % yield, entrapment efficiency and excellent flow properties, optimum friability, good floating property and the drug was released at specific time when symptoms are at peak level in early hours by maintaining required lag phase during floating in acidic medium for 5 hours followed by burst release in simulated intestinal fluid (pH 7.4 phosphate buffer) and followed first order kinetics.
