Formulation Development and Optimization of Taste Masked Mouth Dissolve Tablets of Promethazine Hydrochloride

Introduction: Taste masked mouth dissolve tablets of Promethazine Hydrochloride were formulated by direct compression method using 22 factorial approach for improved acceptability in children. The effect of two independent variables; croscarmellose sodium and neusilin on in vitro disintegration time was studied by optimization technique. Bitter taste of Promethazine hydrochloride was masked by forming complex with ethyl cellulose and PEG 6000. The tablets were evaluated for in vitro disintegration time, in vitro drug release and standard tablet parameters. The validity of polynomial equation was verified manually by designing checkpoint formulations and determining the response. The results of predicted and experimental values gave good correlation. The results of 22 factorial design revealed that croscarmellose sodium and neusilin significantly influence in vitro disintegration time. Taste masked mouth dissolve tablets of Promethazine hydrochloride with improved palatability and acceptability were developed for pediatric population in shortest time and minimum experimental runs using 22 factorial approach.
