A Topical Novel Drug Delivery System for the Treatment of Localized Psoriasis

Abstract: The purpose of the study was to prepare topical niosomal methotrexate urea in chitosan gel for psoriatic patients. A systematic preparation and investigation of combination of 0.25% niosomal methotrexate and 1% urea gel formulation has been prepared to demonstrate its usefulness in psoriasis patients to avoid systemic side effects caused by a powerful immunosuppressant, when taken orally. Urea as a penetration enhancer and adjuvant in the treatment of psoriasis and used in the formulation to prevent hyper proliferation of cells. The methotrexate niosomes were prepared using lipid layer hydration method using spans. The niosomes prepared were incorporated in chitosan gel. The skin irritation study was carried out in healthy human volunteers by RIPT method. The efficacy of the niosomal gel was tested on psoriatic patients by double blind placebo controlled study and measured using Psoriasis Area and Severity Index (PASI) scoring. The niosomal methotrexate urea gel prepared with chitosan exhibited better efficacy determined using PASI scoring compared to placebo and marketed methotrexate gel.
