Abstract: The clinical evaluation of ofloxacin pastes prepared with different hydrophilic polymers (MC, HPMC, HPC, HEC and NaCMC) was carried out to determine the efficacy of prepared formulation in the treatment of periodontitis. The ethical committee of Navodaya Medical College, Raichur approved the clinical trial protocol. The clinical evaluation was carried out under direct supervision of a dentist. The test teeth selected had a probing depth of 6-10mm with bleeding on probing. All patients were evaluated for probing depth (PD), bleeding on probing (BOP), plaque index (PI) and gingival index (GI). In all these tests, patient groups were treated with formulation, formulation + scaling and root planning, scaling and root planning. All the groups showed a similar based line plaque index (1.42 0.25 to 1.46 0.41). However, plaque index values decreased (to1.02 0.25) remarkably with formulation + scaling and root planning groups when compared to other groups. The baseline Gingival index value were between 1.58 0.36-1.61 0.43 for the all groups that reduced more (p0.05) in formulation + scaling and root planning groups (to 0.49 0.10 at 30th day) compared to other groups. The result of BOP indicated that all groups show significant (p0.05) reduction in bleeding. Interestingly, formulation + scaling and root planning groups showed complete elimination of bleeding sites at 30th day of treatment period. The baseline values of PD (6.44±0.75 to 6.47 ±0.90) deceased significantly (p0.05) in all treated groups, however reduction was greater (p0.05) with groups receiving formulation combined with scaling and root planning (1.61±0.29). These results indicated that combined effect of local delivery of ofloxacin with scaling and root planning was more effective in treating periodontitis. It is concluded that adjunctive use of ofloxacin past results in significant benefits in the treatment of periodontitis.
